I think the highest drafted combo would have been Cadillac Williams and Ronnie Brown at Auburn in the early-mid 2000s
 
Bush/White at USC
Jones/McFadden at Arkansas
Some of the combos at Bama or Wisconsin in recent years?

Thurman Thomas/Barry Sanders at Oklahoma State might be the gold standard in modern times.
Charles White/Marcus Allen at USC.
Leonard Fournette/Derrius Guice at LSU was pretty damn good.
Nick Chubb/Sony Michel at Georgia.
Trent Richardson/Mark Ingram was a devastating combo at Alabama.

The 2001 Miami team was also remarkable for having four future recognizable NFL players, two borderline Hall of Famers, and another guy who was a solid pro (Clinton Portis/Frank Gore/Willis McGahee/Najeh Davenport).
 
Alabama had Derrick Henry, T.J. Yeldon, Alvin Kamara and Kenyan Drake all at the same time, though Kamara redshirted that year and then transferred to junior college.

Alabama also had Mark Ingram, Trent Richardson and Eddie Lacy together.

Auburn had William Andrews, Joe Cribbs and James Brooks at the same time for 2-3 years in the late 70s.

Later, Auburn had Bo Jackson, Lionel James, Brent Fullwood, Tommie Agee and Tim Jessie at the same time. All of them played in the NFL (though Jessie only briefly).

Marcus Allen as the blocking fullback for Charles White at USC in 1978-79 was pretty sporty.
