The "Endless Love" scene in Happy Gilmore gets me every time. God, that movie is hilarious.
 
But to answer your question, there is only one correct answer. This isn't really a competition.
 
The word "comedy" probably disqualifies Bad News Bears (the one that never should've been remade). Yes, it's funny through and through, yet it provides a true sense of what America was about circa 1976. One of my top 5 faves, any genre.
